Technical advances in gene sequencing in the past few years have greatly enhanced our ability to detect and understand the role of genetic alterations in musculoskeletal diseases. Though our treatments based upon the new knowledge has not been substantially affected by the recent advances, this is likely to change in the near future. The orthopaedist who stays abreast of these advances will be able to offer better counseling, diagnostic tests and treatment options for patients. The current group of papers in this symposium provides readers with background on how genetic researchers are trying to unravel the genetic
mystery of both common and rare orthopaedic disorders. An earlier study1 found genetic conditions were present in greater than 50% of patients admitted to a pediatric orthopaedic hospital with musculoskeletal disorders. The genes responsible for Mendelian inherited orthopaedic conditions, including many types of skeletal dysplasias, osteogenesis imperfecta, and neurocutaneous syndromes (neurofibromatosis) are known. For other, more common, conditions with complex inheritance patterns, such as scoliosis and osteoarthritis, the identification of genes that influence disease susceptibility has been more elusive.
